ULTRASTRUCTURE OF LYMPHOCYTE
|

Abstract

|
Many authors have studied the ultra structure of the lymphocyte (Bernhard., 1960; and Bessis., 1961; etc.). Animal studies (Clawson et al., 1966; Sorenson., 1964) have contributed to the general knowledge of blood and blood formation. The ultra structure of normal human blood lymphocyte (Goodman et al., 1957; Low and Freeman., 1958; Douglas et al., 1967; and Anderson., 1966), Bone marrow lymphocyte (Movat and Fernado., 1964), Lymph nodes lymphocyte (Marchesi and Gowans., 1903; Han and Song., 1961) has been reported as well as the finding in peripheral blood lymphocyte (Tanaka and Epstein., 1963; Duglaus et al., 1967; Torell. et al., 1963).
The lymphoblast probably originated from a reticular cells it has a rounded nucleus and large reticulated nucleolus. The chromatin has a fine, and more or less uniformly dispersed structure. The cytoplasm contains the same organelles as the lymphocyte. Rough endoplasmic reticulum practically nonexistent in the cytoplasm of small lymphocytes, is represented by a few entirely flattens sacs. The blast cells of acute leukemia have large pale nuclei and a thin of cytoplasm containing many free ribosome, a few canals of RER, round or oval mitochondria, and sometimes a large band of fibrils. The small lymphocyte is characterized by a densely stained nucleus surrounded by a narrow cytoplasm filled with ribosomes and containing up to a golgi membranes and sometimes a small band of fibrills. Other granules are also, but not invariably, found in the lymphocyte cytoplasm.
A particular vacuole may be in the lymphocyte cytoplasm ("the compound vacuole" of Low and Freeman, 1958). It is a clear vacuole limited by a membrane and containing several minute dense vesicles. This is not a specific structure of the lymphocyte. The cytoplasm of lymphocytes also contains dense granules surrounded by a fine osmiophille membrane.
These are undoubtedly the azurophille granule seen in the light microscope. The large and medium lymphocyte are intermediate in structure between the lymphoblast stage and small lymphocyte.
